From Prajapati Nagar, the Aqua Line runs west through Vaishnodevi Square and Ambedkar Square, into central Nagpur at Nagpur Railway Station and Cotton Market, then through Sitabuldi before continuing via Shankar Nagar Square and Dharampeth College to Subhash Nagar and Vasudev Nagar, terminating at Lokmanya Nagar. Sitabuldi is the Aqua Line's interchange with the Orange Line, near Nagpur's central business district.

Rail Vikas Nigam Limited (RVNL) was named the lowest bidder for civil package C-06 of Nagpur Metro's Aqua Line extension on October 16, 2024. The Rs 307.34-crore contract covers 19.4 km across two Aqua Line extensions: Reach 3A (Lokmanya Nagar to Hingna, 6.6 km, 7 stations) and Reach 4A (Prajapati Square to Transport Nagar, 5.5 km, 3 stations).
